CJC-1295 DAC (DAC:GRF) is a synthetic 29 amino acid analog of growth hormone-releasing hormone (GHRH), developed by ConjuChem. It incorporates a Drug Affinity Complex (DAC) that dramatically extends its half-life to approximately 8 days, compared to minutes for native GHRH.
CJC-1295 DAC is a tetrasubstituted version of GHRH 1-29, with substitutions at positions 2, 8, 15, and 27 to improve enzymatic resistance. Its DAC element — an N-epsilon-3-maleimidopropionamide derivative linked at the C-terminus — enables covalent binding to plasma albumin, extending half-life from minutes to 6–8 days.
This mechanism allows sustained, continuous GH secretion, unlike CJC-1295 without DAC (Mod GRF 1-29) which produces more physiological pulsatile release. Both forms show comparable affinity for GHRH receptors.
