NAD+ (Nicotinamide Adenine Dinucleotide) is an endogenous nucleotide considered the central coenzyme of cellular metabolism. It is involved in over 500 enzymatic reactions and its age-related decline is associated with mitochondrial dysfunction and cellular aging.

OVERVIEW

NAD+ acts as a coenzyme in redox reactions (NAD⁺↔NADH), as a substrate for sirtuins (SIRTs) for mitochondrial homeostasis, for PARPs for genomic stability, and for CD38/CD157 for immunoregulation. These three NAD+-dependent enzyme classes may compete for its availability.

Its two key intermediates — nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) — are studied as potential agents of “productive aging” in numerous animal models.

RESEARCH AREAS
Productive aging — NMN exposure over 12 months in normally aging mice: reduced weight gain, increased energy metabolism, improved lipid profile.
Neuroprotection — restoration of mitochondrial functions in nerve and brain cells of aged mice (NMN exposure, 3–12 months).
DNA repair — significant improvement in BER activity, cell viability, and oxidative DNA damage repair after ischemic stress (neuronal cultures, rats).
Cardiac function — cardioprotective action against ischemic injury in murine models (NMN administered 30 min before induced ischemia).
Skeletal function — possible increase in ATP production, reduced inflammation, and improved mitochondrial functions in aged mouse muscles (NMN, 7 days).
